Background
Myeloid differentiation factor 88 (MYD88), a primary differentiation response gene in Myeloid precursors, was discovered in 1990. MYD88 is a universal connexin containing three main structures: N - terminal death domain (DD), intermediate connexin domain (ID), and C - terminal Toll/interleukin-1 receptor domain (TIR). After ligand binding, the cytoplasmic TIR domain of toll - like receptor (TLR) or MyD88 binds to TIR of MyD88.
The most common point mutation in MYD88 is the replacement of leucine (Leu) with proline (Pro) at site 265, known as the MYD88 L265P mutation. A recurrent MYD88 L265P mutation was first identified in diffuse Large B - cell lymphoma (DLBCL). This mutation is rare or absent in PATIENTS with CMB - DLBCL, primary mediastinal B - cell lymphoma, and Burkitt's lymphoma.
Macroglobulinemia of Fahrenheit (WM)/lymphoplasmacytic lymphoma (LPL) is an indolent non - Hodgkin's lymphoma with a low clinical incidence. MyD88 L265P mutation is accounted for 87% of WM.
