Background
Phosphoinositide 3-kinase (PI3K, phospho PI3K) Binds to activated (phosphorylated) protein-Tyr kinases, through its SH2 domain, and acts as an adapter, mediating the association of the p110 catalytic unit to the plasma membrane. Phospho PI3K is necessary for the insulin-stimulated increase in glucose uptake and glycogen synthesis in insulin-sensitive tissues. During insulin stimulation, it also binds to IRS-1. Growth factors and hormones trigger this phosphorylation event, which in turn coordinates cell growth, cell cycle entry, cell migration, and cell survival.

Western blot: Extracts from 293 cells untreated or treated with sorbitol stained with anti - PI3 Kinase p85 / p55 phospho (Tyr467 / Tyr199) antibody.

Immunocytochemical analysis of Phospho - PI3K p85/p55 (Tyr458)/p55 (Tyr199) in NIH/3T3 cells using a Phospho - PI3K p85/p55 (Tyr458)/p55 (Tyr199) polyclonal antibody. The samples were fixed with PFA and permeabilized in 0.1% saponin prior to blocking in 10% serum for 45 minutes at 37°C. The primary antibody was diluted 1:400 and incubated with the sample for 1 hour at 37°C. A Alexa Fluor 594 conjugated goat polyclonal to rabbit IgG (H+L), diluted 1:600 was used as secondary antibody.
