| 背景介绍 | This gene encodes one member of a family of translation repressor proteins. The protein directly interacts with eukaryotic translation initiation factor 4E (eIF4E), which is a limiting component of the multisubunit complex that recruits 40S ribosomal subunits to the 5' end of mRNAs. Interaction of this protein with eIF4E inhibits complex assembly and represses translation. This protein is phosphorylated in response to various signals including UV irradiation and insulin signaling, resulting in its dissociation from eIF4E and activation of mRNA translation. Immunofluorescence: Formalin-fixed MCF7 cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 min and blocked with 3% BSA-PBS for 30 min at RT. Cells were stained with ARG65800 anti-4E-BP1 antibody in 3% BSA-PBS and incubated overnight at 4°C in a hidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at RT in the dark. DAPI was used to stain the cell nuclei (blue).

Immunohistochemistry: Formalin-fixed and Paraffin-embedded Human lung cancer. Antigen retrieval: Heat mediated with Sodium citrate buffer (pH 6.0). The section was then incubated with ARG65800 anti-4E-BP1 antibody at RT and detected using an HRP conjugad compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
