The encoded protein is a component of gap junctions, which are composed of arrays of intercellular channels that provide a route for the diffusion of low molecular weight materials from cell to cell. The encoded protein is the major protein of gap junctions in the heart that are thought to have a crucial role in the synchronized contraction of the heart and in embryonic development. A related intronless pseudogene has been mapped to chromosome 5. Immunofluorescence: Formalin-fixed HeLa cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were stained with ARG66599 anti-Connexin 43 antibody (red) in 3% BSA-PBS and incubated overnight at 4°C. DAPI (blue) for nuclear staining.

Immunohistochemistry: Formalin-fixed and paraffin-embedded Human breast cancer tissue. Antigen Retrieval: Heat mediation was performed in Sodium citrate buffer (pH 6.0). The section was then stained with ARG66599 anti-Connexin 43 antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
