| 背景介绍 | Human platelet membrane glycoprotein Glycoprotein IIb/IIIa (GP IIb/IIIa) is a dominate receptor on the platelet membrane with very high molecular weight (>200kDa). After the platelet is activated, the GP IIb/IIIa receptor complex changes conformation and binds to fibrinogen in blood circulation with high affinity.The activation of platelets is initiated locally when the endothelial layer is wounded and the platelets are exposed to the collagen underneath the endothelium.The fibrinogen aggregates with platelets when binding to the GP IIb/IIIa receptor on platelets, thus primary haemostasis (Platelet clot) is formed. |
| 生物功能 | Integrin alpha-IIb/beta-3 is a receptor for fibronectin, fibrinogen, plasminogen, prothrombin, thrombospondin and vitronectin. It recognizes the sequence R-G-D in a wide array of ligands. It recognizes the sequence H-H-L-G-G-G-A-K-Q-A-G-D-V in fibrinogen gamma chain. Following activation integrin alpha-IIb/beta-3 brings about platelet/platelet interaction through binding of soluble fibrinogen.
