| 背景介绍 | This is an N-ras oncogene encoding a membrane protein that shuttles between the Golgi apparatus and the plasma membrane. This shuttling is regulated through palmitoylation and depalmitoylation by the ZDHHC9-GOLGA7 complex. The encoded protein, which has intrinsic GTPase activity, is activated by a guanine nucleotide-exchange factor and inactivated by a GTPase activating protein. Mutations in this gene have been associated with somatic rectal cancer, follicular thyroid cancer, autoimmune lymphoproliferative syndrome, Noonan syndrome, and juvenile myelomonocytic leukemia. Western blot: 35 µg of Jurkat (A) and MCF7 (B) cell lysates (in RIPA buffer) stained with ARG64621 anti-N-Ras antibody at 0.3 µg/ml (A) and 0.1 µg/ml (B) dilutions and incubated at RT for 1 hour.

Flow Cytometry: Paraformaldehyde-fixed MCF7 cells permeabilized with 0.5% Triton. Cells were stained with ARG64621 anti-N-Ras antibody (blue line) at 10 µg/ml dilution for 1 hour, followed by incubation with Alexa FluorR 488 labelled secondary antibody. IgG control: Unimmunized goat IgG (black line).
