背景介绍The TATA-binding protein (TBP) plays acentral role in the assembly of mosteukaryotic transcription initiation complexes.TBP assembles with other proteins to formunique multimeric complexes for each of thethree different nuclear RNA polymerases.TBP has been cloned from a variety ofspecies and consists of two distinct domains.The C-terminal domain (~180 amino acids)is highly conserved among species,whereas the N-terminal domain variesconsiderably in length and sequence amongdifferent species. The conserved C-terminaldomain contains the DNA-binding region aswell as regions that interact with positive andnegative regulatory proteins. In human TBP,the non-conserved N-terminal domainincludes a stretch of glutamine residuesranging from 26 to 42. Several researchgroups have found an association betweenexpansion of this polyglutamine tract withrare forms of spinocerebellar ataxia andother neurodegenerative diseases including Huntington's disease.
生物功能General transcription factor that functions at the core of the DNA-binding multiprotein factor TFIID. Binding of TFIID to the TATA box is the initial transcriptional step of the pre-initiation complex (PIC), playing a role in the activation of eukaryotic genes transcribed by RNA polymerase II. Component of the transcription factor SL1/TIF-IB complex, which is involved in the assembly of the PIC (preinitiation complex) during RNA polymerase I-dependent transcription. The rate of PIC formation probably is primarily dependent on the rate of association of SL1 with the rDNA promoter. SL1 is involved in stabilization of nucleolar transcription factor 1/UBTF on rDNA.
