Background
Interleukin-11 Receptor Alpha (IL-11RA) is a subunit of the functional receptor for the pleiotropic cytokine IL-11. It forms a signaling complex with glycoprotein 130 (gp130), activating downstream JAK/STAT, MAPK and PI3K pathways. Primarily expressed in epithelial, stromal, hematopoietic and neuronal cells, IL-11RA mediates critical biological functions including tissue regeneration (intestinal mucosa, liver, bone), hematopoietic support (megakaryopoiesis, platelet production) and embryonic development. Interleukin-11 Receptor Alpha (IL-11RA) holds emerging diagnostic and prognostic relevance in fibro-inflammatory diseases and epithelial cancers. Its overexpression in tissues and elevated soluble levels in serum correlate with pathological fibrosis (lung, liver, kidney), serving as a potential biomarker for disease severity and progression. In oncology, upregulated IL-11RA expression is linked to gastrointestinal cancers (colorectal, gastric), breast cancer and metastasis, where it promotes tumor-stroma crosstalk, chemoresistance and poor survival.
