Background
Transcription factor OVO-like zinc finger 2 (OVOL2) contains a single zinc finger domain composed of a tetrad of C2H2 zinc fingers with variable N- and C-terminal extensions that contains intrinsically disordered domains, which is DNA-binding protein that binds to the 5’-G[CT]TGGGG-3’ core sequence. It probably acts as a transcription regulator. OVOL2 plays a critical role in maintaining the integrity of epithelial lineage. It also suppresses cell cycling and terminal differentiation of keratinocytes by directly repressing MYC and NOTCH1. In addition, OVOL2 is important for the correct development of primordial germ cells in embryos.
OVOL2 is expressed in testis, ovary, heart and skeletal muscle of human normal tissue.
OVOL2 is a colorectal tumor suppressor that blocks WNT signaling by facilitating the recruitment of histone deacetylase 1 to the TCF4-β-catenin complex. Strategies to increase levels of OVOL2 might be developed to reduce colorectal tumor progression and metastasis. In addition, OVOL2 is demonstrated to directly repress the expression of several glycolytic genes, blocking the Warburg effect and breast tumor growth and metastasis.
