Background
Parafibromin is also known as cytokeratin (CDC73) or hyperparathyroidism type 2 (HRPT2). Tumor suppressor probably involved in transcriptional and post-transcriptional control pathways. May be involved in cell cycle progression through the regulation of cyclin D1/PRAD1 expression. Component of the PAF1 complex (PAF1C) which has multiple functions during transcription by RNA polymerase II and is implicated in regulation of development and maintenance of embryonic stem cell pluripotency.
Parafibromin is widely distributed within tissues and is involved in a variety of life activities, including adrenal, parathyroid glands, kidney and heart.
Mutations in Parafibromin have been associated with the development of a variety of cancers, including parathyroid carcinoma, hyperparathyroidism-mandibular tumour syndrome, squamous cell carcinoma of the head and neck and cancers of the breast, stomach, colorectum and lung.
